    Book Description

A thorough understanding of cellular and molecular mechanisms involved in the individual expression of toxic effects provides an important tool for assessment of human health risk. New aspects, major advances, and new areas in molecular and cellular biology and toxicology demand updated sources of information to elucidate the functional mechanics of human toxicology. Mechanistic Toxicology: The Molecular Basis of How Chemicals Disrupt Biological Targets, Second Edition retains the accessible format of the original to present the general principles that link xenobiotic-induced toxicity with the molecular pathways that underlie these toxic effects. Extensively illustrated, this book forms a conceptual bridge between multiple events at the molecular level and the determinants of toxicity at the physiological and cellular level. Specific examples of drugs, environmental pollutants, and other chemicals are carefully chosen to illustrate and highlight the fundamental mechanisms of toxicity at different toxicokinetic and toxicodynamic levels. The book includes references and review articles at the end of each chapter, as well as boxed text for relevant review information on biological, biochemical, molecular, and toxicological background. Linking molecular pathways to more general biomedical contexts, the author ensures that the reader is not lost in the details and instead receives a broad understanding of the processes underlying xenobiotic toxicity. New in the Second Edition · Updated chapters · Types of toxic responses · Disruption of signal transduction by xenobiotics · Disruption of mitochondrial function · Novel mechanisms derived from systems toxicology

      Book Description

Sunscreens are universally recommended by dermatologists not only to prevent the immediate effects of overexposure to sunlight but also to prevent skin cancer. While the former goal is immediately evident, the latter remains an unproven hypothesis and is a topic of some controversy. Recent epidemiological studies suggesting a correlation between increased use of sunscreens over the past two decades and the rise in skin cancer have led to the question whether sunscreens applied to skin may be undergoing photoreactions, the effects of which are elaborated many years later. By addressing the key questions, this book advances the field of sunscreen photobiology and provides the reader with an unbiased perspective on this important field.

      Book Description

Fully updated and rewritten by a basic scientist who is also a practicing physician, the third edition of this popular textbook remains comprehensive, authoritative and readable. Taking a receptor-based, target-centered approach, it presents the concepts central to the study of drug action in a logical, mechanistic way grounded on molecular and biochemical principles. Students of pharmacy, chemistry and pharmacology, as well as researchers interested in a better understanding of drug design, will find this book an invaluable resource. Starting with an overview of basic principles, Medicinal Chemistry examines the properties of drug molecules, the characteristics of drug receptors, and the nature of drug-receptor interactions. Then it systematically examines the various families of receptors involved in human disease and drug design. The first three classes of receptors are related to endogenous molecules: neurotransmitters, hormones and immunomodulators. Next, receptors associated with cellular organelles (mitochondria, cell nucleus), endogenous macromolecules (membrane proteins, cytoplasmic enzymes) and exogenous pathogens (viruses, bacteria) are examined. Through this evaluation of receptors, all the main types of human disease and all major categories of drugs are considered. There have been many changes in the third edition, including a new chapter on the immune system. Because of their increasingly prominent role in drug discovery, molecular modeling techniques, high throughput screening, neuropharmacology and genetics/genomics are given much more attention. The chapter on hormonal therapies has been thoroughly updated and re-organized. Emerging enzyme targets in drug design (e.g. kinases, caspases) are discussed, and recent information on voltage-gated and ligand-gated ion channels has been incorporated. The sections on antihypertensive, antiviral, antibacterial, anti-inflammatory, antiarrhythmic, and anticancer drugs, as well as treatments for hyperlipidemia and peptic ulcer, have been substantially expanded. One new feature will enhance the book's appeal to all readers: clinical-molecular interface sections that facilitate understanding of the treatment of human disease at a molecular level.

5 out of 5 stars Better than Goodman and Gilman for pharmacy students.......2002-01-18

I have taken two pharmacological type classes. One was pharmacology and the other was simply med chem. We used Goodman and Gilman in the pharmacology class and its just too dense. It has much information, but in my opinion, its not appropriate for a textbook. I thought that this book, by Nogrady is alot more managable. However, you need to have a background in biochemistry and organic chemistry to understand some of the topics in this book. It does a good job of explaining underlying mechanisms of pharmaceutical actions. Towards the end, it adopts a Goodman and Gilman approach and just starts listing drugs and their mechanism of action. However, Nogrady doesn't go on for 5 pages like Goodman and Gilman do. Its a bit more concise. Anyway, I don't mean to say that Goodman and Gilman is a bad book, quite the contrary, its an amazing book with more information than one could ever possibly want. Its just that if you want to understand pharmacology for a class, and are not browsing the pages for fun over a cup of tea, Nogrady may be the way to go.

          Book Description

Flow cytometry is a sensitive and quantitative platform for the measurement of particle fluorescence. In flow cytometry, the particles in a sample flow in single file through a focused laser beam at rates of hundreds to thousands of particles per second. During the time each particle is in the laser beam, on the order of ten microseconds, one or more fluorescent dyes associated with that particle are excited. The fluorescence emitted from each particle is collected through a microscope objective, spectrally filtered, and detected with photomultiplier tubes. Flow cytometry is uniquely capable of the precise and quantitative molecular analysis of genomic sequence information, interactions between purified biomolecules and cellular function. Combined with automated sample handling for increased sample throughput, these features make flow cytometry a versatile platform with applications at many stages of drug discovery. Traditionally, the particles studied are cells, especially blood cells; flow cytometry is used extensively in immunology. This volume shows how flow cytometry is integrated into modern biotechnology, dealing with issues of throughput, content, sensitivity, and high throughput informatics with applications in genomics, proteomics and protein-protein interactions, drug discovery, vaccine development, plant and reproductive biology, pharmacology and toxicology, cell-cell interactions and protein engineering.

              Book Description

Advances in Molecular Toxicology features the latest advances in all of the subspecialties of the broad area of molecular toxicology. Toxicology is the study of poisons and this series details the study of the molecular basis by which a vast array of agents encountered in the human environment and produced by the human body itself manifest themselves as toxins. Not strictly limited to documenting these examples the series is also concerned with the complex web of chemical and biological events that give rise to toxin-induced symptoms and disease. The new technologies that are being harnessed to analyze and understand these events will also be reviewed by leading workers in the field.

Advances in Molecular Toxicology will report progress in all aspects of these rapidly evolving molecular aspects of toxicology with a view toward detailed elucidation of both progress on the molecular level and on advances in technological approaches employed.

              * Cutting edge reviews by leading workers in the discipline.
              * In depth dissection of molecular aspects of interest to a broad range of scientists, physisicans and any student in the allied disciplines.
              * Leading edge applications of technological innovations in the chemistry, biochemistry and molecular medicine.
