    Book Description

The various cell types have traditionally been recognized and classified according to their appearance in the light microscope following the process of fixing, processing, sectioning, and staining tissues that is known as histology. Classical histology has been augmented by
immunohistochemistry (the use of specific antibodies to stain particular molecular species in situ). Immunohistochemistry has allowed the identification of many more cell types than could be visualized by classical histology, particularly in the immune system and among the scattered hormone-secreting cells of the endocrine system.

This book discusses all aspects of immunohistochemistry and in situ hybridization technologies and the important role they play in reaching a cancer diagnosis. It provides step-by-step instructions on the methods of additional molecular technologies such as DNA microarrays, and microdissection, along with the benefits and limitations of each method. The topics of region-specific gene expression, its role in cancer development and the techniques that assist in the understanding of the molecular basis of disease are relevant and necessary in science today, ensuring a wide audience for this book.

    * The only book available that translates molecular genetics into cancer diagnosis
    * Provides the readers with tools necessary to perform and optimize sensitive, powerful techniques, including immunohistochemistry and fluorescence in situ hybridization, used in tumor diagnosis
    * Written by experts in this field, the book provides theoretical considerations as well as practical approaches to carry out effectively these techniques
    * Offers suggestions, tips, cautions, and guidelines to avoid artifacts and misdiagnosis
    * Introduces new techniques to detect genes and proteins involved in the initiation and progression of cancer
    * Covers the latest developments and a wide range of applications to the detection of antigens and single-copy DNA and RNA
    * Written in a uniform format, each chapter includes Introduction, Materials required, step-by-step detailed Methods, Results, Discussion, and comprehensive up-to-date References

      Book Description

This completely revised and updated edition takes advantage of the many new developments and applications that have occurred in PRINS and in situ PCR technology, including a new fast multicolor PRINS method for identifying human chromosomes, PRINS protocols for in situ detection of unique sequences and point mutation analysis, a new dideoxy PRINS procedure, and in situ PCR and PRINS protocols for plants. The authors describe in detail various applications of PRINS in human (detection of gene deletions in cancer detection of fetal cells in maternal blood, and assessment of aneuploidy in brain tissues and embryos) as well as plant cells. Readily reproducible in situ PCR techniques are also presented for the detection of cytomegalovirus, and for use in combination with microdissection. In situ RT-PCR techniques are also given for use in plant or cancer investigation.

5 out of 5 stars THE book for in situ methodologies.......1999-11-03

With the rapid and on-going development of In Situ hybridisation, a new edition of this book was needed. In Professor Darby the publishers found a very capable editor. He has gathered articles which, while conveying all the protocols you would need to reproduce the techniques in your own laboratory, are surprisingly readable.

Even for the experienced practitioner, there are methods in here that will catch you by surprise. The wealth of illustrations add to the enjoyment, as you gain a very clear idea of what to expect. It is a shame, however, that most are in black and white. The supplement of colour photos (and the page in the professor's own article) demonstrate the results most clearly.

As the summary says, if you are interested in hybridisation - whether of protein, chromosomes or even whole embryos - this is an indispensable manual.

(caveat: probably not a must-by for the general reader)

        Book Description

Ricardo Lloyd, md, phd, provides critical and tutorial surveys of the cutting-edge molecular and cell biologic techniques used today in the analysis of cells and tissues in situ. These molecular-based histologic and morphologic techniques are described in step-by-step detail by hands-on experts, often those who have originated or perfected them. Among the techniques presented are laser capture microdissection, in situ hybridization, FISH, confocal laser scanning microscopy, in situ PCR, and tyramide amplification. Applications include the detection of infectious agents and DNA, the diagnosis of undifferentiated tumors and endocrine lesions, the metaphase analysis of whole blood cultures, and clonality analysis. In its integrated, state-of-the-art approach, Morphology Methods: Cell and Molecular Biology Techniques thoroughly details and applies the rich panoply of significant new cell and molecular biologic technologies to all the major problems confronting morphologic studies and diagnostic pathology today.

            Book Description

This convenient laboratory manual covers the theory and application of all the commonly used FISH procedures for both the research and clinical service laboratory. Readers are provided with the theoretical principles which underlie each procedure, and are then guided through the protocols in a systematic, easy to follow manner. Because FISH procedures are particularly sensitive to background and subtle variations in sample quality, the trouble-shooting sections are invaluable to the beginner interested in developing FISH capabilities in their laboratory. Areas covered include probe production and labelling; FISH gene mapping; murine, fiber, and RNA FISH; CGH and microchips; 3D FISH; SKY and M-FISH; FISH and microarrays; and FISH in clinical cytogenetics. The information is easy to follow and includes a large number of useful reference web sites in addition to a comprehensive and up to date bibliography providing background to all the advanced molecular cytogenetics techniques in current use. This book will be of particular value to scientists, technologists, and students in the areas of molecular pathology, cancer biology, genomics, haematology, oncology, clinical cytogenetics, molecular diagnostics and genetics.

            Book Description

Immunocytochemistry and in situ hybridization are widely used biomedical sciences. They are essential in medical diagnosis and in cell biology research. Affinity labeling is the central goal of the experimental strategy involving a series of techniques in a logical order; from the effects of specimen fixation, through specimen preparation to expose the antigen, to optimizing immunolabeling, to assessing the result and finally to safety considerations. Numerous examples of these techniques in biomedical sciences are included, as well as experimental assays and practical tips. This survey of methods will serve as an invaluable reference source in any laboratory setting (academic, industrial or clinical) involved in research in almost every branch of biology or medicine, as well as in pharmaceutical, biotechnological and clinical applications.

                Book Description

PCR in situ hybridization allows the detection of specific nucleic acid sequences and their distribution in the cell. It combines two powerful techniquesin situ hybridization (ISH), which allows cellular localization of DNA and RNA sequences in cells and tissues, and the polymerase chain reaction (PCR), which allows reproducible amplification of rare nucleic acid sequences. The combined technique and its variants greatly enhance the sensitivity of in situ hybridization and add morphological localization to the sensitivity of PCR. Such techniques have enormous potential for research and diagnosis but problems with reproducibility and reliability are often encountered. This book overcomes these problems by describing the key procedures in step-by-step detail and by providing the essential advice needed for success. Topics include: DNA in situ PCR and DNA PCR in situ hybridization (PCR ISH) for the detection of DNA targets in cells; reverse trancriptase in situ PCR (RT-PCR) and RT-PCR ISH for the detection of RNA targets; and PRINS (primed in situ synthesis) for chromosomal analysis in interphase nuclei and metaphase chromosome spreads. There are further chapters on fixation of tissues for PCR, selective ultraviolet radiation fractionation (SURF), application of in situ PCR to human tissues, applications and modifications of PCR-ISH, and automation of in situ amplification. PCR In Situ Hybridization is a unique and timely collection of well-tested protocols for the amplification of DNA and RNA in cells and tissues, drawing on the accumulated knowledge and experience of leading exponents of these techniques. For each topic covered, the authors provide detailed guidance on the key steps in the protocols, numerous hints and tips for success, and advice on trouble-shooting. PCR In Situ Hybridization will be invaluable to molecular biologists, pathologists, geneticists, and all those seeking to perform in situ analyses of nucleic acid molecules.
